Peptide Reconstitution and Concentration Maths

Research Use Only. Peptides Lab UK supplies research-grade materials for in-vitro laboratory research only. Nothing on this page is medical advice or an instruction for human or animal use, and no product sold on this website is a medicine.

What this page covers

Laboratory protocols often require a lyophilised peptide to be reconstituted to a known concentration. This page explains the arithmetic only: converting between mass, volume and concentration. It does not give, recommend or imply any dose for humans or animals.

The core formula

Concentration (mg/mL) = mass of peptide in the vial (mg) ÷ volume of diluent added (mL).

Worked example: a vial containing 5 mg of lyophilised peptide reconstituted with 2.0 mL of diluent gives 5 ÷ 2.0 = 2.5 mg/mL, which is 2,500 µg/mL.

Converting a required mass to a volume

Volume (mL) = required mass (mg) ÷ concentration (mg/mL). For example, to obtain 0.5 mg of peptide for an in-vitro assay from a 2.5 mg/mL solution: 0.5 ÷ 2.5 = 0.2 mL. Remember 1 mg = 1,000 µg.

Graduation scales

Some laboratory syringes and pipettes are graduated on a 100-unit scale where 100 units equal 1.0 mL, so 1 unit = 0.01 mL. Converting 0.2 mL on such a scale gives 20 units. This is a volume conversion only.

Good laboratory practice

  • Record lot number, vial mass, diluent and volume.
  • Use calibrated volumetric equipment and aseptic technique.
  • Verify the stated vial content against the certificate of analysis (HPLC/MS).
  • Store solutions as specified and within a validated stability window.

Use our peptide calculator to check concentration arithmetic.

UK regulatory status

In the UK, medicines are regulated by the Medicines and Healthcare products Regulatory Agency (MHRA). Research-grade compounds sold by Peptides Lab UK are supplied strictly for laboratory research; they are not medicinal products, are not manufactured to medicinal GMP standards and must not be used in humans or animals.
